Abstract

The presence of pathogenic bacteria in spices represents a risk to public health as a possible cause of food contamination. The aim of this postgraduate dissertation is to investigate the microbial, antioxidant capacity and antimicrobial activity of a particular group of spices. A total of 9 packed and 27 unpacked samples representing different type of spices were randomly collected from different market and bazaars in North Greece. The microbiological analysis was performed to the laboratory immediately after sample collection. All samples were analyzed in duplicate for microbiological analysis and in triplet for chemical analysis requirements. The spices included in this study were 4 samples each of black pepper (Piper nigrum), turmeric (Curcuma longa), ginger (Zingiber officinale), cinnamon (Cinnamomum verum), pimpinella anisum (Illicium verum), macis (Myristica fragrans Houtt.), tsimeni (Trigonella foenum-graecum), saffron (Crocus sativus) and clove (Syzygium aromaticum). Samples were classified in 4 categories: root, flower, xyloid (wooden like) and seed. The sampling sites were Florina, Xanthi and Thessaloniki. Microbiological analysis was performed for the detection and enumeration of pathogenic microorganisms such as Salmonella spp., Shigella spp., Clostridium perfringens, Escherichia coli and Staphylococcus aureus. Also, the samples were tested for the presence of yeasts and fungi. To perform the microbiological analysis, traditional methods of analysis were used. The determination of the antioxidant capacity of the samples was performed using the ABTS and DPPH methods. The FOLIN-CIOLATEU method was used to determine the total phenolic content. The antimicrobial activity of the samples was determined using the DISC DIFFUSION method.
